A mini-tetracistronic RNA was constructed using three hammerhead ribozymes, which recognize a single sequence in its four exons, and two anti-hairpin ribozymes, which recognize a hairpin structure in the exons. The hairpin structure is required for self-cleavage by the ribozymes. The mini-tetracistronic RNA was stabilized by the introduction of a stem-loop structure and thus displayed high Tm values in a solution containing the components necessary for its in vitro transcription. Inclusion of the anti-hairpin ribozyme in the mini-tetracistronic RNA resulted in an enzyme-substrate complex, which is more stable than the enzyme-substrate complex from the single ribozyme sequence in the pre-tetracistronic RNA. This suggests that the secondary structure formed by the ribozyme sequences in the mini-tetracistronic RNA contributes to the increase in enzyme-substrate complex stability. Such an increase in stability is useful for a more accurate in vitro synthesis of mini-tetracistronic RNAs.[12.17\]
