AutoCAD provides two basic input modes for specifying parts to cut. To investigate the mechanism underlying the luteal defect in PCOS, the objective of the present study was to determine whether there was any association between the defect and hyperinsulinaemia or increased intra-ovarian androgen concentrations. Plasma insulin levels and concentrations of intra-ovarian androgens were measured in anovulatory PCOS women and a matched group of normoovulatory women. The expression of intra-ovarian insulin receptors was determined using immunohistochemistry and Western blotting and the expression of insulin-like growth factor-I (IGF-I) receptor and a phosphorylated form of AKT was examined using a phospho-AKT antibody. To determine whether an inhibition of insulin signaling is involved in the pathogenesis of PCOS, the effect of insulin on cell proliferation was investigated in vitro using an immortalized human luteal cell line, CL2/DCCs. The results showed that PCOS women had lower plasma insulin levels than normoovulatory women. This is not attributable to hyperinsulinaemia, because the concentration of insulin receptors was lower in PCOS women compared with normoovulatory women. Although the intra-ovarian androgen concentrations were also lower, this decrease was not directly attributable to increased androgen production, as the expression of 5alpha-reductase was similar in PCOS and normoovulatory women.
