In this project we propose to determine whether the glycoproteins are synthesized in the cytoplasm and if so whether they are targeted to the erythrocyte surface through a mechanism similar to that found in secretion. If such a targeting process exists for the glycoproteins, we will determine whether it is specific for the sialoglycoproteins or whether it is a more generalized phenomenon. If the biosynthesis of these macromolecules in the cytoplasm is sufficient to account for their surface expression on the erythrocyte we will seek to identify those aspects of the sialoglycoprotein synthesis which control the process of surface expression. In this process, we will also address the question of whether the sialoglycoproteins are initially synthesized in a form that allows them to reach the cell surface but, upon maturation of the sialic acid residues, these proteins are no longer properly expressed at the surface. Specific antibodies will be employed to define the characteristics of their cellular distribution, their synthesis and their processing by sialytransferase in tissue culture cells. As a final aim, we will determine whether these sialoglycoproteins are associated with a binding site for calcium, a modification which may be requisite for their subsequent insertion into the membrane. As a first approximation of the roles of these glycoproteins in erythrocyte function, we will determine whether their removal by sialidase alters the membrane’s permeability to water and to ions. Such an alteration may be responsible for the defects in the erythrocyte which may account for increased solute exchange in sickle cell anemia and other diseases in which this enzyme activity is high.President Donald Trump’s threat to shut down the government over his demand for billions of dollars to build a wall on the Mexican border may backfire in the end, some Republicans predict. Democrats said the meeting had no clear plan to avert a possible shutdown. Mark Sanford of South Carolina made the comment. “I learned in high school that the expectation was that the government would fail to keep its business on the assumption that it would force the congress, or parts of it, to back off in spending,” Sanford said on “CBS This Morning.” “I know the president from my wilderness years in North Carolina when we were shut down. So, I’m sure that he learned that lesson.
